Description
L1TD1 (LINE-1 Type Transposase Domain Containing 1) is a protein-coding gene that encodes a protein containing a domain homologous to the LINE-1 retrotransposon open reading frame 1 (ORF1) protein. It is highly expressed in embryonic stem cells and is considered a marker of pluripotency. L1TD1 is involved in maintaining stem cell identity and self-renewal, and its expression is frequently dysregulated in various cancers, including germ cell tumors and somatic malignancies.

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations that truncate the protein are predicted to cause loss of function, impairing stem cell maintenance.
Gain of Function (GOF)
No gain-of-function mutations have been reported in L1TD1.
Dominant Negative (DN)
No dominant-negative mutations have been characterized for L1TD1.

Protein Summary
The L1TD1 protein is a 413-amino acid polypeptide that contains a coiled-coil domain and a domain similar to the LINE-1 ORF1 protein. It localizes to both the nucleus and cytoplasm and is thought to bind RNA and proteins involved in stem cell pluripotency. L1TD1 interacts with key pluripotency factors such as NANOG and OCT4, and its expression is essential for maintaining the undifferentiated state of embryonic stem cells.
