Description

KLRG2 (killer cell lectin like receptor G2) is a protein-coding gene located on chromosome 7q34. It encodes a transmembrane receptor belonging to the C-type lectin family, expressed primarily on natural killer (NK) cells and a subset of T cells. KLRG2 functions as an inhibitory receptor, binding to cadherins to regulate immune cell activation and cytotoxicity. It plays a role in immune surveillance, tumor immunity, and autoimmune regulation.

Protein Summary

KLRG2 is a type II transmembrane glycoprotein with an extracellular C-type lectin-like domain. It forms disulfide-linked homodimers and binds to classical cadherins (e.g., E-cadherin, N-cadherin). Upon ligand binding, KLRG2 recruits phosphatases via its immunoreceptor tyrosine-based inhibitory motif (ITIM)-like sequence, leading to inhibition of NK cell and T cell effector functions. It is considered an immune checkpoint receptor.
