Description

KLK3 (kallikrein-related peptidase 3) encodes prostate-specific antigen (PSA), a serine protease primarily produced by the prostate epithelium. PSA is the most widely used clinical biomarker for prostate cancer screening, diagnosis, and monitoring. The protein is secreted into seminal plasma where it liquefies the semen coagulum. KLK3 expression is regulated by androgens and is highly tissue-specific, with minimal expression outside the prostate. Clinically, elevated serum PSA levels correlate with prostate pathology including benign prostatic hyperplasia (BPH), prostatitis, and prostate cancer. The gene is part of the kallikrein gene cluster on chromosome 19.

Protein Summary

Prostate-specific antigen (PSA) is a 261-amino-acid glycoprotein synthesized as a preproenzyme. After cleavage of the signal peptide and pro-peptide, the mature active enzyme (237 amino acids) is secreted. PSA is a chymotrypsin-like serine protease that cleaves semenogelin I and II in seminal coagulum, facilitating sperm motility. In serum, PSA exists in free and complexed forms (bound to alpha-1-antichymotrypsin). The protein has a single catalytic triad (His57, Asp102, Ser195) and is stabilized by five disulfide bonds. PSA is used clinically as a biomarker; its half-life in serum is approximately 2-3 days.
