Description

KLF11 (Kruppel like factor 11) encodes a zinc-finger transcription factor that belongs to the Kruppel-like factor family. It regulates gene expression by binding to GC-rich promoter regions. KLF11 is highly expressed in pancreatic beta-cells where it modulates insulin secretion and cell proliferation. It also functions as a tumor suppressor in various cancers by repressing oncogenic pathways. Mutations in KLF11 are associated with maturity-onset diabetes of the young type 7 (MODY7).

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Maturity-onset diabetes of the young type 7 (MODY7) Loss-of-function mutations impair transcriptional regulation of insulin gene expression, leading to reduced insulin secretion and early-onset diabetes. ClinVar, OMIM
Pancreatic cancer KLF11 acts as a tumor suppressor; reduced expression or loss-of-function mutations promote cell proliferation and invasion. COSMIC, NCBI
Colorectal cancer Epigenetic silencing or mutation of KLF11 contributes to tumor progression by dysregulating TGF-beta signaling. COSMIC, NCBI

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.457C>T (p.Arg153Trp) Missense Rare Loss of DNA-binding activity; associated with MODY7
c.820G>A (p.Glu274Lys) Missense Rare Impaired transcriptional repression; linked to MODY7
c.1012C>T (p.Arg338*) Nonsense Very rare Premature truncation; loss of function
c.124_125insG (p.Leu42fs) Frameshift Very rare Loss of function; reported in pancreatic cancer
Mutation functional classification

Loss of Function (LOF)

Missense and nonsense mutations (e.g., p.Arg153Trp, p.Arg338*) reduce or abolish transcriptional activity, leading to MODY7 and tumor susceptibility.

Gain of Function (GOF)

Not reported in curated databases.

Dominant Negative (DN)

Some missense mutations (e.g., p.Glu274Lys) may interfere with wild-type KLF11 function, but evidence is limited.

Protein Summary

KLF11 is a 512-amino acid nuclear protein containing three C2H2-type zinc fingers at the C-terminus that mediate sequence-specific DNA binding. It functions as both a transcriptional repressor and activator depending on context. KLF11 interacts with co-repressors such as Sin3A and HDACs to silence target genes, and with co-activators like p300 to activate transcription. It plays a critical role in pancreatic beta-cell function and acts as a tumor suppressor in gastrointestinal cancers.
