Description

KIF24 is a member of the kinesin superfamily of microtubule-based motor proteins. It plays a critical role in the regulation of ciliogenesis by depolymerizing microtubules at the mother centriole, thereby suppressing primary cilia formation in cycling cells. KIF24 is also implicated in cell cycle progression and has been associated with various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (general) Dysregulation of KIF24 may promote uncontrolled cell proliferation by altering cilia-dependent signaling pathways. COSMIC; literature review
Breast cancer Overexpression of KIF24 has been observed in breast cancer cell lines, potentially contributing to tumorigenesis. COSMIC; NCBI Gene
Lung cancer KIF24 mutations and altered expression reported in lung cancer samples. COSMIC

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1234C>T (p.Arg412*) Nonsense <0.1% Premature truncation; likely loss of function
c.567G>A (p.Val189Met) Missense <0.1% Unknown significance; predicted benign
c.890_891insA Frameshift <0.1% Frameshift; likely loss of function
Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations (e.g., p.Arg412*, c.890_891insA) are predicted to result in loss of KIF24 function, potentially disrupting cilia regulation.

Gain of Function (GOF)

No documented gain-of-function mutations in KIF24.

Dominant Negative (DN)

No evidence for dominant-negative effects in KIF24.

Pathways

• Cilium assembly (Reactome: R-HSA-5617833)
• Microtubule-dependent transport (Reactome: R-HSA-983189)

Protein Summary

KIF24 encodes a kinesin motor protein that localizes to the mother centriole and uses its ATPase activity to depolymerize microtubules, thereby preventing primary cilium formation in proliferating cells. The protein contains an N-terminal motor domain and a C-terminal domain involved in centriolar targeting. KIF24 is essential for cell cycle-dependent cilia disassembly and its dysregulation is linked to cancer.
