Description
KIF14 (kinesin family member 14) encodes a microtubule-based motor protein belonging to the kinesin-3 family. It is essential for cytokinesis, mitotic spindle assembly, and chromosome segregation. KIF14 interacts with citron kinase and localizes to the central spindle and midbody. Loss-of-function mutations cause Meckel syndrome type 12 (MKS12), a severe ciliopathy. Overexpression of KIF14 is observed in multiple cancers and is associated with poor prognosis, likely due to chromosomal instability.

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations (e.g., p.Arg76*) lead to truncated protein, loss of motor activity, and cause Meckel syndrome 12.
Gain of Function (GOF)
Overexpression and certain missense variants (e.g., p.Glu1094Lys) may enhance microtubule binding or processivity, contributing to cancer cell proliferation.
Dominant Negative (DN)
Not clearly established; some missense variants may interfere with wild-type KIF14 function in cytokinesis.

Protein Summary
KIF14 is a 1,648-amino acid kinesin-3 family motor protein with an N-terminal motor domain, a central coiled-coil stalk, and a C-terminal tail. It moves along microtubules toward the plus end, transporting cargoes essential for cytokinesis and ciliogenesis. The protein localizes to the central spindle and midbody during mitosis, interacting with citron kinase to ensure proper abscission. In cilia, KIF14 is required for intraflagellar transport and Hedgehog signaling. Dysregulation leads to ciliopathy or cancer.
