Description

ING5 (Inhibitor of Growth Family Member 5) is a protein-coding gene that encodes a tumor suppressor protein. It is a component of the HBO1 (MYST1) and MOZ/MORF (MYST3/MYST4) histone acetyltransferase complexes, which acetylate histones H3 and H4 to regulate chromatin structure and gene expression. ING5 is involved in cell cycle arrest, DNA repair, and apoptosis. Its expression is frequently downregulated in various cancers, and mutations or deletions contribute to tumorigenesis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Loss of ING5 expression reduces HBO1 complex activity, impairing DNA repair and promoting genomic instability. PMID: 21499277, COSMIC
Colorectal cancer Decreased ING5 mRNA and protein levels correlate with poor prognosis; ING5 overexpression induces apoptosis. PMID: 21804532, NCBI Gene
Hepatocellular carcinoma ING5 downregulation promotes cell proliferation and invasion via altered histone acetylation. PMID: 25362251, ClinVar
Gastric cancer ING5 promoter hypermethylation leads to silencing and tumor progression. PMID: 23959598, OMIM
Lung cancer Reduced ING5 expression associated with advanced stage and metastasis. PMID: 22664980, COSMIC

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G (p.Met1?) Missense <0.1% Likely loss of start codon; predicted loss of function
c.205C>T (p.Arg69Trp) Missense <0.1% Found in breast cancer; disrupts PHD finger domain
c.421_422delAG (p.Ser141fs) Frameshift deletion <0.1% Truncating; loss of nuclear localization signal
c.634C>T (p.Arg212*) Nonsense <0.1% Premature stop; nonsense-mediated decay expected
Mutation functional classification

Loss of Function (LOF)

Most reported ING5 mutations (nonsense, frameshift, missense in PHD domain) lead to loss of histone binding and reduced HAT complex activity, impairing tumor suppression.

Gain of Function (GOF)

No gain-of-function mutations have been reported for ING5.

Dominant Negative (DN)

No dominant-negative mutations have been characterized; however, truncated forms could potentially interfere with wild-type ING5 function.

Pathways

Histone acetylation by HBO1 complex (Reactome: R-HSA-3214847)
Chromatin modifying enzymes (KEGG: hsa05203)
p53 signaling pathway (KEGG: hsa04115)
DNA damage response (Reactome: R-HSA-73894)

Protein Summary

The ING5 protein (UniProt Q8N3W0) is a 240-amino-acid nuclear protein containing a conserved PHD finger domain that binds to histone H3 trimethylated at lysine 4 (H3K4me3). It is a core component of the HBO1 and MOZ/MORF histone acetyltransferase complexes, which acetylate histones H3 and H4 to promote open chromatin and gene activation. ING5 regulates cell cycle progression, DNA repair, and apoptosis. Loss of ING5 function contributes to cancer development through epigenetic dysregulation.
