Description

The ING3 gene encodes a member of the inhibitor of growth (ING) family of tumor suppressors. The protein contains a plant homeodomain (PHD) finger, which binds to histone H3 trimethylated at lysine 4 (H3K4me3), and is a component of the NuA4 histone acetyltransferase complex. ING3 functions in chromatin remodeling, transcriptional regulation, cell cycle arrest, and apoptosis. It is frequently downregulated or mutated in various cancers, suggesting a role in tumor suppression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Head and neck squamous cell carcinoma Loss of ING3 expression leads to reduced apoptosis and increased proliferation PMID: 18369447; COSMIC
Melanoma Decreased ING3 expression correlates with tumor progression and poor prognosis PMID: 17909063; ClinVar
Breast cancer ING3 downregulation associated with aggressive tumor phenotypes PMID: 21351256; NCBI Gene
Colorectal cancer Reduced ING3 expression linked to metastasis and worse survival PMID: 23175147; COSMIC

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.497C>T (p.Pro166Leu) Missense 0.02% (gnomAD) Unknown; may affect PHD finger function
c.103G>A (p.Gly35Arg) Missense 0.01% (gnomAD) Unknown; located in N-terminal region
c.1A>G (p.Met1?) Start loss <0.01% Likely loss of function
Mutation functional classification

Loss of Function (LOF)

Frameshift or nonsense mutations leading to truncated protein or loss of PHD finger are predicted to impair chromatin binding and tumor suppressor activity.

Gain of Function (GOF)

No gain-of-function mutations have been reported for ING3.

Dominant Negative (DN)

Missense mutations in the PHD finger may disrupt histone binding and exert dominant-negative effects, but evidence is limited.

Pathways

• NuA4 histone acetyltransferase complex
• p53 signaling pathway
• Apoptosis
• Cell cycle - G1/S transition

Protein Summary

ING3 is a 47 kDa nuclear protein containing a PHD finger that specifically recognizes trimethylated histone H3 at lysine 4 (H3K4me3). It is a stable component of the NuA4/Tip60 histone acetyltransferase complex, which acetylates histones and non-histone proteins such as p53. Through this complex, ING3 regulates transcription of genes involved in cell cycle arrest, DNA repair, and apoptosis. Loss of ING3 expression or function contributes to tumorigenesis in multiple cancer types.
