Description

The ING1 (inhibitor of growth family member 1) gene encodes a tumor suppressor protein that regulates cell cycle progression, apoptosis, and chromatin remodeling. It is a member of the ING family of proteins, which contain a plant homeodomain (PHD) finger that binds to histone H3 trimethylated at lysine 4 (H3K4me3). ING1 interacts with the p53 signaling pathway and is frequently downregulated or mutated in various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Loss of ING1 expression leads to reduced p53-mediated apoptosis and increased cell proliferation. PMID: 10655551, COSMIC
Head and neck squamous cell carcinoma Decreased ING1 expression correlates with tumor progression and poor prognosis. PMID: 11994725
Melanoma ING1 mutations and loss of heterozygosity contribute to tumorigenesis. PMID: 12592314
Gastric cancer Reduced ING1 expression is associated with advanced tumor stage and lymph node metastasis. PMID: 15042678
Lung cancer ING1 downregulation promotes cell cycle progression and resistance to apoptosis. PMID: 15586223

Mutation functional classification

Loss of Function (LOF)

Most ING1 mutations result in loss of tumor suppressor activity, including impaired p53 interaction and reduced apoptosis.

Gain of Function (GOF)

No gain-of-function mutations have been reported for ING1.

Dominant Negative (DN)

Some missense mutations (e.g., p.Arg166Gln) may act in a dominant-negative manner by competing with wild-type ING1 for binding partners.

Pathways

• p53 signaling pathway (KEGG: hsa04115)
• Apoptosis (KEGG: hsa04210)
• Cell cycle (KEGG: hsa04110)

Protein Summary

The ING1 protein (p33ING1) is a 33 kDa nuclear protein containing a PHD finger domain that specifically recognizes histone H3 trimethylated at lysine 4 (H3K4me3). It functions as a chromatin reader and recruits histone acetyltransferase (HAT) complexes to promote gene expression. ING1 enhances p53 acetylation and stability, thereby inducing cell cycle arrest and apoptosis in response to DNA damage. It also interacts with PCNA and is involved in DNA repair. Loss of ING1 expression or function contributes to tumorigenesis in multiple cancer types.
