Description

IL31 (interleukin 31) is a cytokine gene located on chromosome 12q24.31. It encodes a 164-amino acid protein that belongs to the IL-6 cytokine family. IL31 signals through a heterodimeric receptor composed of IL31RA and OSMR. It is primarily produced by activated Th2 cells and plays a key role in pruritus (itching), inflammation, and immune regulation. Dysregulation of IL31 is implicated in atopic dermatitis, prurigo nodularis, and other inflammatory skin conditions.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Atopic Dermatitis IL31 overexpression promotes Th2 inflammation and pruritus via IL31RA signaling in sensory neurons. NCBI Gene, OMIM
Prurigo Nodularis Elevated IL31 levels correlate with chronic itch and nodular skin lesions. NCBI Gene, OMIM
Allergic Contact Dermatitis IL31 contributes to itch and inflammation in allergen-exposed skin. NCBI Gene

Protein Summary

Interleukin 31 (IL-31) is a 164-amino acid cytokine with a four-helix bundle structure. It is secreted by activated Th2 cells and binds to the IL31RA/OSMR receptor complex, activating JAK-STAT, PI3K/AKT, and MAPK pathways. IL-31 induces pruritus by directly stimulating sensory neurons and promotes inflammation by upregulating chemokines and cytokines in keratinocytes and immune cells.
