Description

IL24 (Interleukin 24) is a member of the IL-10 family of cytokines. It is primarily expressed in immune cells and skin, and functions as a tumor suppressor by inducing apoptosis selectively in cancer cells. IL24 also plays roles in wound healing, inflammation, and immune regulation. The gene is located on chromosome 1q32.1 and encodes a secreted protein that signals through IL-20 receptor complexes.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Melanoma IL24 induces apoptosis via endoplasmic reticulum stress and mitochondrial pathways; loss of IL24 expression contributes to tumor progression. PMID: 11526407; NCBI Gene
Breast Cancer IL24 overexpression suppresses tumor growth and metastasis through anti-angiogenic and pro-apoptotic mechanisms. PMID: 15569927; COSMIC
Psoriasis IL24 is upregulated in psoriatic skin lesions and may contribute to inflammation and keratinocyte hyperproliferation. PMID: 12640457; OMIM
Rheumatoid Arthritis IL24 levels are elevated in synovial fluid and serum, promoting inflammatory cytokine production. PMID: 19116917; UniProt

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations in IL24 are predicted to cause loss of tumor suppressor activity, potentially contributing to cancer progression.

Gain of Function (GOF)

No gain-of-function mutations have been reported in IL24.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for IL24.

Pathways

IL-10 family signaling pathway (Reactome: R-HSA-6783783)
Apoptosis (KEGG: hsa04210)
JAK-STAT signaling pathway (KEGG: hsa04630)

Protein Summary

IL24 is a 206-amino acid secreted cytokine with a signal peptide (residues 1-48) and a mature region (49-206). It contains an IL-10 family signature domain. The protein binds to IL-20R1/IL-20R2 or IL-22R1/IL-20R2 receptor complexes, activating STAT3 signaling. IL24 selectively induces apoptosis in cancer cells via endoplasmic reticulum stress and reactive oxygen species production, while sparing normal cells. It also modulates immune responses and angiogenesis.
