Description
IL22 (Interleukin 22) is a member of the IL-10 family of cytokines. It is produced by Th17 cells, Th22 cells, NK cells, and innate lymphoid cells. IL22 signals through a heterodimeric receptor composed of IL22RA1 and IL10RB, activating STAT3 and MAPK pathways. It plays key roles in mucosal immunity, tissue repair, and antimicrobial defense, particularly in epithelial cells of the skin, intestine, and respiratory tract. Dysregulation of IL22 is implicated in inflammatory diseases such as psoriasis, inflammatory bowel disease (IBD), and certain cancers.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Psoriasis | IL22 overexpression drives keratinocyte hyperproliferation and inflammation via STAT3 activation. | PMID: 19234213, 24958822 |
| Inflammatory Bowel Disease (IBD) | IL22 promotes epithelial barrier integrity and antimicrobial peptide production; dysregulation contributes to chronic inflammation. | PMID: 18641395, 23543747 |
| Rheumatoid Arthritis | IL22 levels elevated in synovial fluid; promotes osteoclastogenesis and joint inflammation. | PMID: 21593343 |
| Gastric Cancer | IL22 signaling via STAT3 promotes tumor cell proliferation and survival. | PMID: 24714568 |
| Colorectal Cancer | IL22 can promote tumorigenesis through STAT3-mediated proliferation and anti-apoptotic signals. | PMID: 22895138 |

Pathways
• IL-22 signaling pathway (Reactome: R-HSA-8984722)
• JAK-STAT signaling pathway (KEGG: hsa04630)
• Cytokine-cytokine receptor interaction (KEGG: hsa04060)
Protein Summary
IL22 is a 179-amino-acid cytokine (UniProt Q9GZX6) with a signal peptide (residues 1-33) and a mature chain (34-179). It adopts a typical alpha-helical cytokine fold. IL22 binds to the IL22RA1/IL10RB receptor complex, activating STAT3, MAPK, and PI3K/AKT pathways. It is primarily produced by immune cells and acts on non-hematopoietic epithelial cells to promote antimicrobial defense, tissue repair, and barrier integrity.
