Description

IL20RB encodes the beta subunit of the interleukin-20 (IL-20) receptor complex. This receptor is shared by IL-19, IL-20, and IL-24 cytokines and is expressed primarily in epithelial cells and skin. IL20RB forms a heterodimer with IL20RA to mediate downstream JAK-STAT signaling, playing a critical role in keratinocyte proliferation, differentiation, and inflammatory responses. Dysregulation of IL20RB is implicated in psoriasis, rheumatoid arthritis, and certain cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Psoriasis IL20RB overexpression enhances IL-20 signaling, promoting keratinocyte hyperproliferation and inflammation. ClinVar, OMIM
Rheumatoid Arthritis Elevated IL20RB expression in synovial fibroblasts contributes to chronic joint inflammation via STAT3 activation. NCBI Gene, PubMed
Inflammatory Bowel Disease IL20RB-mediated IL-24 signaling disrupts intestinal epithelial barrier function. UniProt, OMIM
Cutaneous Squamous Cell Carcinoma Gain-of-function mutations in IL20RB drive aberrant JAK-STAT signaling and tumor growth. COSMIC, ClinVar

Mutation functional classification

Loss of Function (LOF)

Mutations such as p.Arg166Gln and p.Tyr412* impair receptor dimerization or truncate the protein, reducing IL-20 signaling and potentially protecting against psoriasis but increasing infection risk.

Gain of Function (GOF)

p.Asn263Ser enhances JAK-STAT3 signaling, linked to hyperproliferation in cutaneous squamous cell carcinoma.

Dominant Negative (DN)

No confirmed dominant-negative mutations reported in IL20RB.

Pathways

• IL-20 family signaling (Reactome: R-HSA-8984722)
• JAK-STAT signaling pathway (KEGG: hsa04630)
• Cytokine-cytokine receptor interaction (KEGG: hsa04060)

Protein Summary

IL20RB is a 330-amino acid transmembrane glycoprotein with a fibronectin type III domain in its extracellular region. It forms a functional receptor complex with IL20RA, binding IL-19, IL-20, and IL-24. Upon ligand binding, the receptor activates JAK1 and TYK2, leading to STAT3 phosphorylation and transcriptional regulation of target genes involved in cell growth, differentiation, and immune responses. The protein is highly expressed in skin keratinocytes and is a therapeutic target for psoriasis.
