Description
IL1F10 encodes interleukin-38 (IL-38), a member of the interleukin-1 cytokine family. IL-38 binds to the IL-1 receptor accessory protein (IL1RAP) and the IL-36 receptor (IL-36R), functioning as an anti-inflammatory cytokine by inhibiting IL-36 signaling. It is expressed in skin, tonsils, and certain immune cells, and has been implicated in inflammatory and autoimmune diseases.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Rheumatoid arthritis | IL-38 reduces IL-36-mediated inflammation by blocking IL-36R signaling | PMID: 25687188 |
| Psoriasis | IL-38 downregulation in psoriatic skin leads to increased IL-36-driven inflammation | PMID: 26014625 |
| Systemic lupus erythematosus | IL-38 levels correlate with disease activity and modulate cytokine production | PMID: 28264816 |

Protein Summary
IL-38 is a 152-amino acid protein with a typical IL-1 family beta-trefoil fold. It lacks a signal peptide and is secreted via a non-classical pathway. IL-38 binds to IL-36R and IL1RAP, antagonizing IL-36-induced NF-κB and MAPK activation, thereby suppressing pro-inflammatory cytokine production.
