Description

IL13RA1 encodes a subunit of the interleukin-13 receptor complex. This protein binds IL13 with low affinity but, upon heterodimerization with IL4R, forms a high-affinity receptor that mediates IL13 signaling. IL13RA1 is involved in allergic inflammation, asthma, fibrosis, and certain cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Asthma IL13RA1 forms a receptor complex with IL4R to transduce IL13 signals, promoting airway hyperresponsiveness and mucus production. ClinVar, OMIM
Allergic rhinitis Enhanced IL13 signaling via IL13RA1 contributes to IgE production and eosinophilic inflammation. OMIM
Pulmonary fibrosis IL13RA1-mediated signaling drives fibroblast activation and collagen deposition. NCBI Gene, OMIM
Hodgkin lymphoma IL13RA1 overexpression supports autocrine IL13 growth loops in Reed-Sternberg cells. COSMIC, NCBI Gene
Glioblastoma IL13RA1 is overexpressed in glioblastoma; targeted therapies using IL13-PE38 immunotoxin are under investigation. COSMIC, NCBI Gene

Protein Summary

IL13RA1 is a 427-amino acid transmembrane glycoprotein (UniProt P78552) with an extracellular domain containing a fibronectin type III domain. It binds IL13 with low affinity and, upon association with IL4R, forms a functional receptor that activates JAK1/TYK2 and STAT6. The protein is expressed in immune cells, lung, and various tumors. It is a target for therapeutic antibodies (e.g., lebrikizumab) and immunotoxins in cancer.
