Description

IGFBP6 (Insulin Like Growth Factor Binding Protein 6) encodes a member of the insulin-like growth factor binding protein (IGFBP) family. This protein has a high affinity for insulin-like growth factor II (IGF-II) and modulates IGF-II signaling by sequestering it in the extracellular space, thereby inhibiting its mitogenic and anti-apoptotic effects. IGFBP6 is expressed in various tissues and is implicated in cell growth, differentiation, and apoptosis. It is also involved in the regulation of cancer cell proliferation and metastasis.

Protein Summary

IGFBP6 is a 240-amino acid secreted protein (UniProt P24592) with a molecular weight of approximately 27 kDa. It contains an N-terminal IGFBP domain and a C-terminal thyroglobulin type-1 domain. The protein binds preferentially to IGF-II over IGF-I, with high affinity, and inhibits IGF-II-mediated signaling by preventing its interaction with the IGF1 receptor. IGFBP6 also has IGF-independent effects, including modulation of cell adhesion and migration. It is expressed in many tissues and is often downregulated in cancers, where its loss contributes to tumor progression.
