Description

IFNA5 encodes a member of the type I interferon family, interferon alpha-5, which is secreted by immune cells in response to viral infection. This cytokine binds to the IFNAR receptor complex, activating JAK-STAT signaling to induce an antiviral state, inhibit cell proliferation, and modulate immune responses. IFNA5 is part of a cluster of interferon alpha genes on chromosome 9p21.3.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hepatitis C virus infection Reduced IFNA5 expression impairs antiviral response; therapeutic IFN-alpha used ClinVar, PMID: 25428589
Systemic lupus erythematosus Dysregulated type I interferon signaling including IFNA5 contributes to autoimmunity OMIM, PMID: 29739914
Chronic myeloid leukemia IFNA5 is part of interferon-based therapy; response linked to genetic variants COSMIC, PMID: 21844182

Protein Summary

Interferon alpha-5 is a 189-amino-acid secreted cytokine with a signal peptide (residues 1-23) and a mature chain (24-189). It forms a five-helix bundle typical of type I interferons. The protein binds to the IFNAR1/IFNAR2 receptor complex, initiating phosphorylation of STAT1 and STAT2, leading to transcription of interferon-stimulated genes (ISGs). IFNA5 exhibits antiviral, antiproliferative, and immunomodulatory activities.
