Description
IFITM1 (Interferon Induced Transmembrane Protein 1) is a member of the interferon-induced transmembrane protein family. It is encoded by a gene located on chromosome 11p15.5. The protein is a small transmembrane protein that localizes to the plasma membrane and endosomal compartments. IFITM1 is strongly induced by type I and type II interferons and plays a critical role in the innate immune response by restricting the entry of a broad range of enveloped viruses, including influenza A virus, dengue virus, and SARS-CoV-2. Beyond antiviral activity, IFITM1 is involved in cell proliferation, cell adhesion, and immune cell signaling. Dysregulation of IFITM1 expression has been implicated in various cancers, where it can act as either an oncogene or a tumor suppressor depending on the context.

Protein Summary
IFITM1 is a 125-amino acid transmembrane protein with two hydrophobic domains and a conserved intracellular loop. It is localized to the plasma membrane and endosomes. The protein inhibits viral entry by blocking membrane fusion, likely by altering membrane fluidity and curvature. IFITM1 also modulates cell proliferation and adhesion through interactions with CD81 and other tetraspanins. Post-translational modifications include palmitoylation and ubiquitination, which regulate its stability and antiviral activity.
