Description

IFIT3 encodes a member of the interferon-induced tetratricopeptide repeat (IFIT) family. The protein contains multiple tetratricopeptide repeats that mediate protein-protein interactions. IFIT3 is strongly induced by type I interferons and plays a critical role in antiviral innate immunity by inhibiting viral replication and translation. It also modulates cellular signaling pathways, including NF-κB and IRF3, and is implicated in cancer progression and autoimmune diseases.

Pathways

• Antiviral mechanism by IFN-stimulated genes (Reactome: R-HSA-1169410)
• RIG-I/MDA5 mediated induction of IFN-alpha/beta (Reactome: R-HSA-168928)

Protein Summary

IFIT3 is a 56 kDa protein with multiple tetratricopeptide repeats that facilitate interactions with viral proteins and host factors. It inhibits viral translation by binding to the eukaryotic initiation factor 3 (eIF3) complex and sequestering viral RNA. IFIT3 also enhances interferon signaling by stabilizing IRF3 and promoting NF-κB activation. Its expression is tightly regulated by interferons and is often dysregulated in cancers and autoimmune disorders.
