Description

IFIT1B (Interferon Induced Protein with Tetratricopeptide Repeats 1B) is a member of the IFIT family of interferon-stimulated genes. It encodes a protein containing multiple tetratricopeptide repeats (TPR) that mediate protein-protein interactions. IFIT1B is induced by type I interferons and plays a role in the innate immune response by recognizing and binding viral RNA, thereby inhibiting viral replication and translation. It is closely related to IFIT1 (ISG56) but has distinct RNA-binding specificity.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Viral infections (e.g., influenza, flavivirus) IFIT1B binds viral RNA lacking 2'-O-methylation, inhibiting viral translation and replication Experimental evidence from cell-based assays and animal models
Systemic lupus erythematosus (SLE) Dysregulated interferon signaling may alter IFIT1B expression, contributing to autoimmune pathology Association studies and transcriptomic profiling

Protein Summary

The IFIT1B protein (UniProt Q5VZK9) contains multiple tetratricopeptide repeats (TPR) that form a superhelical structure for binding viral RNA. It specifically recognizes RNA lacking 2'-O-methylation at the 5' cap, a feature of many viral transcripts. By sequestering viral RNA, IFIT1B inhibits translation initiation and suppresses viral replication. The protein is predominantly cytoplasmic and is rapidly induced upon interferon stimulation.
