Description
HSPA1B encodes a 70 kDa heat shock protein that is strongly induced by cellular stress (e.g., heat, oxidative stress, inflammation). As a molecular chaperone, it assists in protein folding, prevents aggregation of denatured proteins, and facilitates protein transport and degradation. It also plays roles in immune responses and cytoprotection. HSPA1B is part of the highly homologous HSPA1A/HSPA1B pair located in the major histocompatibility complex (MHC) class III region.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Rheumatoid Arthritis | HSPA1B polymorphisms (e.g., +1267 A/G) may influence susceptibility and severity, possibly via altered chaperone function and immune modulation. | Association studies; PMID: 10984555 |
| Type 1 Diabetes | Variants in HSPA1B have been linked to risk, potentially through effects on antigen presentation and autoimmune response. | Genetic association; PMID: 15199473 |
| Alzheimer's Disease | HSPA1B expression is altered in affected brains; chaperone dysfunction may contribute to protein aggregation pathology. | Expression studies; PMID: 16936722 |
| Parkinson's Disease | HSPA1B may modulate alpha-synuclein toxicity; polymorphisms could affect disease risk. | Functional studies; PMID: 20624804 |

Mutation functional classification
Loss of Function (LOF)
Rare loss-of-function mutations are not well documented; complete knockout in mice is viable but shows impaired stress tolerance.
Gain of Function (GOF)
No clear gain-of-function mutations reported; overexpression is generally protective.

Protein Summary
HSPA1B is a 70 kDa heat shock protein that functions as an ATP-dependent molecular chaperone. It binds to exposed hydrophobic patches of unfolded or misfolded proteins, promoting proper folding and preventing aggregation. It also participates in the degradation of irreversibly damaged proteins via the ubiquitin-proteasome system. In addition to its chaperone role, HSPA1B modulates immune responses by influencing antigen presentation and cytokine production. Its expression is low under normal conditions but rapidly induced upon stress, making it a key cytoprotective factor.
