Description

GPX7 encodes glutathione peroxidase 7, a selenium-independent peroxidase that reduces hydrogen peroxide and organic hydroperoxides. It functions as a sensor of oxidative stress and plays a role in protecting cells from oxidative damage. GPX7 is also implicated in tumor suppression, particularly in esophageal adenocarcinoma, where loss of expression correlates with poor prognosis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Esophageal Adenocarcinoma Loss of GPX7 expression leads to increased oxidative stress and DNA damage, promoting tumorigenesis. PMID: 23431130, PMID: 25670082
Barrett's Esophagus Reduced GPX7 levels are associated with progression from Barrett's metaplasia to adenocarcinoma. PMID: 25670082
Hepatocellular Carcinoma GPX7 downregulation contributes to oxidative stress and malignant transformation. PMID: 28723891
Breast Cancer GPX7 hypermethylation and reduced expression are observed in breast cancer cell lines. PMID: 21573163

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G (p.Met1?) Missense <0.1% Likely loss of start codon; predicted loss of function
c.202C>T (p.Arg68Trp) Missense <0.1% Located in catalytic domain; may impair peroxidase activity
c.344G>A (p.Arg115His) Missense <0.1% Uncertain significance; reported in ClinVar
Mutation functional classification

Loss of Function (LOF)

Loss-of-function mutations (e.g., start codon loss, catalytic domain missense) reduce peroxidase activity, leading to increased oxidative stress and potential tumor promotion.

Gain of Function (GOF)

No gain-of-function mutations have been reported for GPX7.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for GPX7.

Pathways

• Glutathione metabolism (Reactome: R-HSA-174403)
• Detoxification of Reactive Oxygen Species (Reactome: R-HSA-3299685)
• Oxidative Stress Induced Senescence (WikiPathways: WP3875)

Protein Summary

GPX7 is a 187-amino acid protein (molecular weight ~21 kDa) localized to the cytoplasm and endoplasmic reticulum. It contains a conserved thioredoxin-like fold and a catalytic cysteine residue (Cys57) instead of selenocysteine, distinguishing it from selenium-dependent GPX family members. GPX7 reduces H2O2 and phospholipid hydroperoxides, protecting cells from oxidative damage. It also interacts with other redox proteins such as PRDX6 and TXN. Loss of GPX7 expression is a hallmark of several cancers, particularly esophageal adenocarcinoma.
