Description

GPR18 (G protein-coupled receptor 18) is a protein-coding gene located on chromosome 13q32.3. It encodes a rhodopsin-like GPCR that is activated by N-arachidonyl glycine (NAGly) and related lipid mediators. GPR18 is involved in immune regulation, cell migration, and cannabinoid signaling. It is expressed in lymphoid tissues, brain, and reproductive organs. Mutations and altered expression have been implicated in inflammatory diseases and cancer.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Inflammatory bowel disease Altered GPR18 signaling may modulate immune cell trafficking and cytokine release PMID: 25687263
Prostate cancer GPR18 expression is upregulated and may promote cell proliferation via MAPK pathway PMID: 23960097
Endometriosis GPR18 expression in endometrial tissue may influence lesion growth and inflammation PMID: 25687263

Protein Summary

GPR18 is a 331-amino acid G protein-coupled receptor with seven transmembrane domains. It is activated by N-arachidonyl glycine (NAGly) and related lipids, leading to Gi/o-mediated signaling, inhibition of cAMP, and activation of MAPK pathways. The receptor is expressed in immune cells, brain, and reproductive tissues, and plays roles in chemotaxis, cell proliferation, and inflammation. Structural studies are limited; functional data derive from heterologous expression systems.
