Description

GPR158 is an orphan G protein-coupled receptor (GPCR) that is highly expressed in the brain, particularly in the hippocampus and cortex. It plays a role in neuronal signaling, synaptic plasticity, and has been implicated in cancer progression, including prostate and breast cancer. GPR158 is known to interact with RGS7 and regulate G protein signaling.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Prostate cancer Overexpression of GPR158 promotes cell proliferation and invasion through GPCR signaling pathways. PMID: 25977332
Breast cancer GPR158 is upregulated and associated with poor prognosis; may activate MAPK/ERK signaling. PMID: 30518678
Major depressive disorder Genetic variants in GPR158 are associated with altered stress response and depression risk. PMID: 28472653

Protein Summary

GPR158 is a 7-transmembrane domain orphan GPCR of approximately 120 kDa. It is predominantly expressed in the brain and forms a complex with RGS7 and Gβ5 to modulate G protein signaling. The protein is involved in neuronal development and synaptic function. In cancer, GPR158 overexpression correlates with aggressive disease and may serve as a therapeutic target.
