Description

GPR146 is an orphan G protein-coupled receptor (GPCR) that is highly conserved across vertebrates. It is implicated in lipid metabolism and cholesterol homeostasis. Genome-wide association studies have linked variants near GPR146 to plasma lipid levels and cardiovascular risk. The receptor is expressed in multiple tissues, including liver and adipose, and may modulate hepatic very low-density lipoprotein (VLDL) secretion.

Protein Summary

GPR146 is a 333-amino acid orphan GPCR with seven transmembrane domains. It is predominantly expressed in liver and adipose tissue. The receptor is thought to couple to Gαi/o proteins, inhibiting adenylyl cyclase and reducing cAMP levels. Functional studies suggest GPR146 regulates hepatic VLDL secretion, thereby influencing plasma triglyceride and cholesterol levels. Its endogenous ligand remains unknown.
