Description

GPC5 (glypican 5) is a member of the glypican family of heparan sulfate proteoglycans, which are attached to the cell surface via a glycosylphosphatidylinositol (GPI) anchor. Glypicans play roles in cell growth, differentiation, and migration by modulating signaling pathways such as Wnt, Hedgehog, and fibroblast growth factors. GPC5 is implicated in several cancers and developmental disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Altered GPC5 expression may influence Wnt signaling and tumor progression PMID: 25691885 (NCBI)
Lung cancer GPC5 polymorphisms associated with susceptibility; overexpression in non-small cell lung cancer PMID: 20676098 (NCBI)
Ovarian cancer GPC5 copy number gains and overexpression linked to poor prognosis PMID: 23593055 (NCBI)
Simpson-Golabi-Behmel syndrome (SGBS) GPC5 mutations may contribute to the phenotype, though GPC3 is primary OMIM 600352

Mutation functional classification

Loss of Function (LOF)

Nonsense mutations (p.Arg412*, p.Trp189*) lead to truncated, non-functional protein.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported.

Dominant Negative (DN)

No evidence for dominant-negative effects.

Pathways

• Wnt signaling pathway (Reactome: R-HSA-195721)
• Hedgehog signaling pathway (Reactome: R-HSA-5358351)
• Glypican pathway (Reactome: R-HSA-1650814)

Protein Summary

Glypican-5 (UniProt P78333) is a 572-amino acid heparan sulfate proteoglycan anchored to the plasma membrane via GPI. It carries heparan sulfate chains that interact with growth factors, morphogens, and extracellular matrix components. GPC5 modulates signaling pathways critical for development and is dysregulated in various cancers.
