Description
GLP2R encodes the glucagon-like peptide 2 receptor (GLP-2R), a G protein-coupled receptor (GPCR) that binds glucagon-like peptide-2 (GLP-2). GLP-2R is primarily expressed in the gastrointestinal tract and plays a critical role in promoting intestinal epithelial cell proliferation, nutrient absorption, and mucosal integrity. Activation of GLP-2R stimulates cAMP production and downstream signaling pathways that regulate intestinal growth and function.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Short bowel syndrome | GLP-2R activation by exogenous GLP-2 (teduglutide) enhances intestinal adaptation and absorption | Clinical trials and FDA approval |
| Crohn disease | GLP-2R signaling may reduce inflammation and promote mucosal healing | Observational studies and clinical trials |
| Intestinal failure | GLP-2R agonists improve nutrient absorption and reduce parenteral nutrition dependence | Clinical evidence |

Mutation functional classification
Loss of Function (LOF)
Mutations that disrupt receptor expression or GLP-2 binding, such as start codon loss, are predicted to cause loss of function.
Gain of Function (GOF)
No gain-of-function mutations have been reported in GLP2R.
Dominant Negative (DN)
No dominant-negative mutations have been described for GLP2R.

Protein Summary
The GLP-2 receptor is a 553-amino acid protein with seven transmembrane domains characteristic of class B GPCRs. It is activated by GLP-2, leading to Gs protein coupling and increased intracellular cAMP. The receptor is essential for mediating the intestinotrophic effects of GLP-2, including stimulation of crypt cell proliferation, inhibition of apoptosis, and enhancement of nutrient transport.
