Description
GKN2 (gastrokine 2) is a stomach-specific gene encoding a secreted protein involved in maintaining gastric mucosal integrity and suppressing gastric carcinogenesis. It is frequently downregulated in gastric cancer and Helicobacter pylori infection.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Gastric Cancer | Loss of GKN2 expression contributes to tumor progression and metastasis | Multiple studies show reduced GKN2 mRNA and protein in gastric tumors vs. normal tissue (PMID: 17981184, 21573172) |
| Helicobacter pylori Infection | H. pylori infection downregulates GKN2, impairing mucosal defense | Observed in gastric biopsies (PMID: 19030185) |

Protein Summary
Gastrokine 2 is a 184-amino acid secreted protein expressed predominantly in gastric mucosa. It forms a complex with gastrokine 1 (GKN1) and trefoil factor 1 (TFF1), protecting the gastric epithelium from injury and suppressing inflammation-driven carcinogenesis.
