Description

GCNT3 encodes a member of the beta-1,6-N-acetylglucosaminyltransferase family. This enzyme catalyzes the transfer of N-acetylglucosamine (GlcNAc) to the core 1 O-glycan structure (Galβ1-3GalNAcα1-Ser/Thr) to form core 2 O-glycans (GlcNAcβ1-6(Galβ1-3)GalNAcα1-Ser/Thr). It is specifically expressed in mucin-secreting tissues and plays a critical role in the biosynthesis of mucin-type O-glycans, influencing cell adhesion, signaling, and tumor progression.

Protein Summary

The GCNT3 protein (UniProt O95395) is a type II transmembrane glycosyltransferase localized to the Golgi apparatus. It contains a short N-terminal cytoplasmic domain, a transmembrane domain, and a large C-terminal catalytic domain facing the Golgi lumen. The enzyme uses UDP-GlcNAc as a donor substrate to add GlcNAc in a beta-1,6 linkage to the core 1 O-glycan structure, generating core 2 O-glycans. This modification is essential for the formation of complex mucin-type O-glycans that modulate cell-cell interactions, pathogen binding, and tumor progression.
