Description
GBP2 (Guanylate Binding Protein 2) is an interferon-inducible member of the dynamin superfamily of large GTPases. It is involved in innate immune responses against intracellular pathogens, regulation of inflammation, and modulation of cell proliferation. GBP2 exhibits antiviral and antibacterial activity, and its expression is upregulated by type I and type II interferons. The protein localizes to the cytoplasm and can associate with membranes, contributing to vesicle trafficking and pathogen restriction.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Infectious diseases (e.g., influenza, HIV, Mycobacterium tuberculosis) | GBP2 restricts viral and bacterial replication through GTPase-dependent mechanisms and inflammasome activation. | NCBI Gene, UniProt |
| Cancer (e.g., breast, colorectal, lung) | GBP2 expression is altered in various cancers; it may act as a tumor suppressor or promoter depending on context, influencing cell migration and immune evasion. | COSMIC, NCBI Gene |
| Inflammatory disorders | GBP2 modulates NLRP3 inflammasome assembly and IL-1β secretion, contributing to chronic inflammation. | UniProt, OMIM |

Protein Summary
GBP2 is a 67 kDa interferon-inducible GTPase that belongs to the dynamin superfamily. It hydrolyzes GTP to GDP and GMP, and is involved in antimicrobial and antiviral immunity. The protein promotes inflammasome activation, vesicle trafficking, and pathogen restriction. It is expressed in immune tissues and upregulated by interferons. Structural studies reveal a large GTPase domain and a helical bundle that mediates membrane association.
