Description

GAS6 (Growth Arrest Specific 6) encodes a vitamin K-dependent protein that acts as a ligand for TAM receptor tyrosine kinases (TYRO3, AXL, MERTK). It is involved in cell survival, proliferation, migration, and phagocytosis. GAS6 is structurally similar to protein S but lacks anticoagulant activity. It plays roles in cancer progression, autoimmune diseases, thrombosis, and neurodegenerative disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Systemic lupus erythematosus GAS6 deficiency impairs clearance of apoptotic cells via TAM receptors, leading to autoantigen exposure and immune activation. ClinVar, OMIM
Venous thromboembolism GAS6 variants may alter plasma levels and affect coagulation regulation. ClinVar, NCBI
Non-small cell lung cancer GAS6 overexpression activates AXL signaling, promoting tumor cell survival and metastasis. COSMIC, NCBI
Glioblastoma GAS6-AXL signaling contributes to therapy resistance and tumor invasion. COSMIC, NCBI
Acute ischemic stroke Elevated GAS6 levels associated with poor outcome and larger infarct size. ClinVar, NCBI

Mutation functional classification

Loss of Function (LOF)

Mutations that reduce GAS6 secretion or receptor binding, impairing apoptotic cell clearance and immune homeostasis.

Gain of Function (GOF)

Overexpression or activating mutations that enhance AXL signaling, promoting cancer cell survival and metastasis.

Dominant Negative (DN)

Not well documented; rare variants may interfere with wild-type GAS6 function in dimerization.

Pathways

• TAM receptor signaling pathway (Reactome: R-HSA-8939211)
• Apoptosis and survival (KEGG: hsa04210)
• Phagocytosis (Reactome: R-HSA-2029480)
• Vitamin K metabolism (Reactome: R-HSA-6803544)

Protein Summary

GAS6 is a 75 kDa vitamin K-dependent glycoprotein composed of an N-terminal Gla domain, four EGF-like domains, and a C-terminal sex hormone-binding globulin (SHBG) domain. The Gla domain mediates binding to phosphatidylserine on apoptotic cells, while the SHBG domain binds TAM receptors. GAS6 is secreted and circulates in plasma. It is a key regulator of efferocytosis, cell survival, and immune tolerance.
