Description

The GAK gene encodes cyclin G associated kinase, a serine/threonine kinase that plays a critical role in clathrin-mediated endocytosis by acting as a co-chaperone for Hsc70. GAK is also involved in cell cycle regulation through its interaction with cyclin G and CDK5. Mutations in GAK have been associated with Parkinson disease and cancer.

Protein Summary

GAK (cyclin G associated kinase) is a 160 kDa protein containing an N-terminal serine/threonine kinase domain, a central J-domain (DnaJ) that recruits Hsc70, and a C-terminal clathrin-binding domain. It functions as an auxilin-like co-chaperone essential for uncoating clathrin-coated vesicles during endocytosis. GAK also phosphorylates cyclin G and interacts with CDK5, linking endocytosis to cell cycle control. The protein is ubiquitously expressed, with highest levels in brain and testis.
