Description

FZD7 encodes a member of the Frizzled family of seven-transmembrane domain receptors that function as receptors for Wnt signaling proteins. FZD7 plays a critical role in embryonic development, cell polarity, and tissue homeostasis. It is frequently overexpressed in various cancers and is implicated in cancer stem cell maintenance and metastasis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer FZD7 overexpression activates canonical Wnt/β-catenin signaling, promoting tumor growth and invasion. High expression correlates with poor prognosis (PMID: 25944712)
Hepatocellular Carcinoma FZD7 upregulation enhances Wnt/β-catenin signaling, contributing to cancer stem cell self-renewal. Overexpression in HCC tissues vs. normal liver (PMID: 20010866)
Breast Cancer FZD7 mediates Wnt signaling to promote epithelial-mesenchymal transition (EMT) and metastasis. High FZD7 expression in triple-negative breast cancer (PMID: 25016929)
Gastric Cancer FZD7 activates β-catenin/TCF transcription, driving proliferation and invasion. FZD7 knockdown reduces tumor growth in xenografts (PMID: 23934182)
Ovarian Cancer FZD7 overexpression correlates with chemoresistance and poor survival. Upregulated in ovarian cancer cell lines (PMID: 21804532)

Protein Summary

FZD7 is a 574-amino acid seven-transmembrane receptor that binds Wnt ligands, particularly Wnt3a and Wnt5a. It activates both canonical (β-catenin-dependent) and non-canonical Wnt signaling pathways. The protein contains a cysteine-rich domain (CRD) in its extracellular N-terminus essential for Wnt binding. FZD7 is highly conserved across species and is a key regulator of cell proliferation, differentiation, and migration. Its overexpression in multiple cancers makes it a potential therapeutic target.
