Description

FZD5 encodes a member of the frizzled family of seven-transmembrane domain receptors that function as receptors for WNT signaling proteins. FZD5 specifically binds WNT5A and WNT3A, activating canonical (β-catenin-dependent) and non-canonical pathways. It plays critical roles in embryonic development, cell polarity, and tissue homeostasis. Aberrant FZD5 expression and mutations are implicated in colorectal cancer, hepatocellular carcinoma, and other malignancies.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer FZD5 overexpression activates β-catenin/TCF transcription, promoting cell proliferation PMID: 19029980
Hepatocellular Carcinoma FZD5 upregulation correlates with WNT/β-catenin pathway activation and poor prognosis PMID: 21252315
Retinoblastoma FZD5 mutations (e.g., p.Gly488Glu) alter WNT signaling in tumorigenesis PMID: 25231345

Protein Summary

Frizzled-5 (FZD5) is a 585-amino acid seven-transmembrane receptor with an N-terminal cysteine-rich domain (CRD) essential for WNT ligand binding. It localizes to the plasma membrane and transduces signals via Dishevelled (DVL) to β-catenin stabilization or planar cell polarity pathways. Post-translational modifications include N-glycosylation and palmitoylation. FZD5 is highly expressed in testis, colon, and liver, and its dysregulation contributes to oncogenic WNT signaling.
