Description
FZD10 encodes Frizzled-10, a seven-transmembrane domain receptor for Wnt signaling proteins. It is a member of the Frizzled family and plays a role in embryonic development, cell polarity, and proliferation. FZD10 is implicated in several cancers, including colorectal, breast, and synovial sarcoma, where it is often overexpressed and associated with poor prognosis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Colorectal Cancer | Overexpression of FZD10 activates canonical Wnt/β-catenin signaling, promoting tumor growth and metastasis. | PubMed: 20406972, COSMIC |
| Synovial Sarcoma | FZD10 is highly expressed in synovial sarcoma cells and contributes to oncogenic Wnt signaling. | PubMed: 15604238, COSMIC |
| Breast Cancer | FZD10 upregulation correlates with aggressive subtypes and poor survival via Wnt pathway activation. | PubMed: 24803677, COSMIC |
| Gastric Cancer | FZD10 overexpression enhances cell proliferation and invasion through Wnt/β-catenin signaling. | PubMed: 27323851, COSMIC |

Mutation functional classification
Loss of Function (LOF)
Frameshift mutations (e.g., p.Thr563Asnfs*12) are predicted to truncate the C-terminal domain, impairing receptor signaling.
Gain of Function (GOF)
No confirmed gain-of-function mutations reported in COSMIC or ClinVar.
Dominant Negative (DN)
No dominant-negative mutations described for FZD10.

Protein Summary
Frizzled-10 (UniProt Q9ULW2) is a 581-amino acid seven-transmembrane receptor with an N-terminal cysteine-rich domain (CRD) essential for Wnt ligand binding. It localizes to the plasma membrane and transduces signals via Dishevelled to stabilize β-catenin. FZD10 is a potential therapeutic target in cancers where it is aberrantly expressed.
