Description

FPGT encodes fucose-1-phosphate guanylyltransferase, an enzyme that catalyzes the conversion of fucose-1-phosphate and GTP to GDP-L-fucose and diphosphate. GDP-L-fucose is a key substrate for fucosyltransferases in the synthesis of fucosylated glycoproteins and glycolipids, which are critical for cell adhesion, immune response, and development. Mutations in FPGT are associated with congenital disorders of glycosylation.

Mutation functional classification

Loss of Function (LOF)

Missense and nonsense variants that reduce or abolish fucose-1-phosphate guanylyltransferase activity, leading to GDP-L-fucose deficiency.

Gain of Function (GOF)

No gain-of-function mutations reported.

Dominant Negative (DN)

No dominant-negative mutations reported.

• Fucose metabolism
• Glycosylation reactions

Protein Summary

Fucose-1-phosphate guanylyltransferase is a 591-amino acid protein localized to the cytosol and Golgi. It catalyzes the reversible conversion of fucose-1-phosphate and GTP to GDP-L-fucose and diphosphate. GDP-L-fucose is the essential donor substrate for fucosyltransferases that modify glycoproteins and glycolipids. The enzyme is highly expressed in liver and kidney, and its deficiency leads to a congenital disorder of glycosylation characterized by impaired fucosylation.
