Aliases TYK3, c-FER, FER (fps/fes related) tyrosine kinase

Description

The FER gene encodes a member of the FPS/FES family of non-receptor tyrosine kinases. This protein is involved in the regulation of cell-cell adhesion, cell migration, and signal transduction pathways, including those mediated by growth factors and cytokines. FER is implicated in various cancers and developmental processes.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer FER overexpression promotes cell migration and invasion via STAT3 signaling PMID: 23431177
Colorectal cancer FER amplification and activation correlate with poor prognosis and metastasis PMID: 25944712
Prostate cancer FER kinase activity enhances androgen receptor signaling and tumor growth PMID: 27325648

Pathways

• PI3K-Akt signaling pathway (KEGG: hsa04151)
• Rap1 signaling pathway (KEGG: hsa04015)
• Focal adhesion (KEGG: hsa04510)
• EGFR tyrosine kinase inhibitor resistance (KEGG: hsa01521)

Protein Summary

FER is a 822-amino acid non-receptor tyrosine kinase containing an N-terminal FCH domain, a central SH2 domain, and a C-terminal tyrosine kinase domain. It localizes to the cytoplasm and plasma membrane, where it phosphorylates substrates involved in cell adhesion, migration, and survival. FER is widely expressed in hematopoietic and epithelial tissues.
