Description

FCGR2C encodes a member of the Fc gamma receptor family (FcγRIIc, CD32c) that binds the Fc region of immunoglobulin G (IgG). This receptor is expressed on natural killer (NK) cells and some myeloid cells, mediating antibody-dependent cellular cytotoxicity (ADCC) and immune complex clearance. The gene is located in the FCGR gene cluster on chromosome 1q23.3 and exhibits copy number variation and polymorphisms associated with autoimmune diseases and infection susceptibility.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Systemic lupus erythematosus (SLE) FCGR2C copy number variation alters IgG immune complex clearance, promoting autoantibody deposition and inflammation. OMIM 604590; PMID 24905358
Rheumatoid arthritis Polymorphisms in FCGR2C affect FcγRIIc expression on NK cells, modulating ADCC and joint inflammation. OMIM 604590; PMID 23028350
Kawasaki disease FCGR2C variants influence IgG receptor function, contributing to coronary artery vasculitis. OMIM 604590; PMID 21841781
Idiopathic thrombocytopenic purpura (ITP) Altered FcγRIIc expression on NK cells impairs platelet clearance regulation. OMIM 604590; PMID 19786223

Loss of Function (LOF)

Nonsense variant rs114945036 introduces premature stop codon, leading to truncated non-functional FcγRIIc.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in FCGR2C.

Dominant Negative (DN)

No dominant-negative mutations described for FCGR2C.

Pathways

Fc gamma R-mediated phagocytosis (KEGG hsa04666)

Protein Summary

Fc gamma receptor IIc (CD32c) is a transmembrane glycoprotein of approximately 40 kDa that binds IgG immune complexes. It contains two extracellular Ig-like domains and a cytoplasmic immunoreceptor tyrosine-based activation motif (ITAM). Expressed primarily on NK cells, it triggers ADCC and cytokine release upon IgG engagement. Alternative splicing and copy number variation contribute to functional diversity and disease susceptibility.
