Description

FCER1G encodes the gamma subunit of the high-affinity IgE receptor (FcεRI), a tetrameric complex (αβγ2) expressed on mast cells and basophils. The gamma subunit contains an immunoreceptor tyrosine-based activation motif (ITAM) that initiates downstream signaling upon IgE cross-linking, leading to degranulation, cytokine release, and allergic inflammation. FCER1G also associates with other Fc receptors (FcγRI, FcγRIIIa) and the T-cell receptor complex, broadening its role in immune regulation.

Mutation functional classification

Loss of Function (LOF)

Mutations in the start codon or ITAM domain (e.g., p.Met1?, p.Arg28Trp) impair receptor signaling and mast cell activation.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in FCER1G.

Dominant Negative (DN)

Deletion in transmembrane domain (p.Lys69del) may exert dominant-negative effects by disrupting receptor complex formation.

Pathways

Fc epsilon RI signaling pathway (KEGG: hsa04664)
Fc gamma R-mediated phagocytosis (KEGG: hsa04666)
Natural killer cell mediated cytotoxicity (KEGG: hsa04650)

Protein Summary

FCER1G encodes the gamma subunit (FcRγ) of the high-affinity IgE receptor. This 86-amino-acid protein contains a single ITAM motif essential for signal transduction. FcRγ dimerizes with the FcεRI alpha and beta subunits to form the functional receptor on mast cells and basophils. Upon IgE cross-linking, ITAM tyrosines are phosphorylated by Src family kinases, recruiting Syk kinase and initiating downstream pathways (MAPK, PI3K, calcium flux) that lead to degranulation and cytokine production. FcRγ also pairs with FcγRI, FcγRIIIa, and the T-cell receptor CD3 complex, highlighting its pleiotropic role in immune signaling.
