Description
FBXO4 encodes a member of the F-box protein family, which is characterized by an approximately 40-amino acid F-box motif. The protein is a component of the SCF (SKP1-CUL1-F-box protein) ubiquitin ligase complex, which mediates ubiquitination and subsequent proteasomal degradation of target proteins. FBXO4 specifically recognizes and binds to phosphorylated cyclin D1, targeting it for degradation, thereby regulating cell cycle progression. It also interacts with other substrates involved in DNA replication and repair. Loss of FBXO4 function has been implicated in various cancers, including breast, lung, and colorectal cancers.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Breast cancer | FBXO4 loss leads to cyclin D1 accumulation, promoting cell cycle progression and tumorigenesis | PMID: 16325584; COSMIC |
| Lung cancer | Reduced FBXO4 expression correlates with poor prognosis and increased proliferation | PMID: 21804531; NCBI Gene |
| Colorectal cancer | FBXO4 mutations impair ubiquitination of cyclin D1, contributing to genomic instability | PMID: 21930794; COSMIC |
| Melanoma | FBXO4 downregulation associated with increased cyclin D1 levels and tumor growth | PMID: 22037554; ClinVar |

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations (e.g., p.Arg339*, p.Leu497fs) result in truncated or non-functional protein, impairing cyclin D1 degradation and promoting cell proliferation.
Gain of Function (GOF)
No gain-of-function mutations have been reported for FBXO4.
Dominant Negative (DN)
Missense mutations (e.g., p.Arg416Gln) may act in a dominant-negative manner by competing with wild-type FBXO4 for substrate binding without promoting ubiquitination.

Protein Summary
FBXO4 is a 497-amino acid F-box protein that serves as the substrate recognition component of the SCF ubiquitin ligase complex. It contains an N-terminal F-box domain (residues 1-40) that binds SKP1, and a C-terminal substrate-binding domain that recognizes phosphorylated cyclin D1. The protein is predominantly cytoplasmic and regulates G1/S cell cycle transition. Post-translational modifications include phosphorylation, which modulates its stability and activity.
