Description

FBXO28 encodes a member of the F-box protein family, characterized by an F-box motif. F-box proteins constitute one of the four subunits of the SCF (SKP1-cullin-F-box) ubiquitin ligase complex, which targets proteins for ubiquitination and subsequent proteasomal degradation. FBXO28 specifically recognizes and binds phosphorylated substrates, playing a critical role in cell cycle progression, DNA damage response, and transcriptional regulation. It has been implicated in the regulation of MYC stability and activity.

• Ubiquitin-mediated proteolysis (KEGG: hsa04120)
• MYC transcriptional regulation (Reactome: R-HSA-212436)

Protein Summary

FBXO28 is a 469-amino acid F-box protein that functions as the substrate recognition component of the SCF ubiquitin ligase complex. It contains an N-terminal F-box domain (residues 1-40) that mediates binding to SKP1, and a C-terminal substrate-binding domain. FBXO28 targets phosphorylated proteins for ubiquitination, with key substrates including MYC. By regulating MYC stability and transcriptional activity, FBXO28 influences cell proliferation, differentiation, and tumorigenesis. The protein is predominantly nuclear and is expressed in multiple tissues, with highest levels in testis and lymphoid organs.
