Description
EXOC8 encodes a component of the exocyst complex, an octameric protein complex essential for tethering secretory vesicles to the plasma membrane prior to fusion. The exocyst complex is involved in exocytosis, cell polarity, and ciliogenesis. EXOC8 is widely expressed and plays a role in neuronal development and function.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Neurodevelopmental disorder with microcephaly, seizures, and brain atrophy | Loss-of-function mutations in EXOC8 disrupt exocyst complex assembly, impairing vesicle trafficking and neuronal development. | ClinVar, OMIM |
| Joubert syndrome (related ciliopathy) | EXOC8 mutations may affect ciliary function via disrupted exocyst-mediated vesicle transport to cilia. | OMIM, PubMed |

Protein Summary
EXOC8 is a 75 kDa protein that localizes to the plasma membrane and vesicle membranes. It interacts with other exocyst subunits (e.g., EXOC1, EXOC2) and small GTPases such as RAB11 and RAL to regulate vesicle tethering. The protein contains a pleckstrin homology (PH) domain and coiled-coil regions important for complex assembly.
