ERG Gene (ETS Transcription Factor)
ERG (ETS-related gene) is a key regulator of hematopoiesis and vascular development, frequently implicated in prostate cancer and Ewing sarcoma through gene fusions.

Description
ERG (ETS-related gene) encodes a member of the ETS family of transcription factors. It is involved in embryonic development, cell proliferation, differentiation, and angiogenesis. ERG is a critical regulator of hematopoiesis and vascular integrity. Chromosomal rearrangements involving ERG, most notably TMPRSS2-ERG fusion, are common in prostate cancer. ERG fusions are also characteristic of Ewing sarcoma and acute myeloid leukemia. Overexpression and mutations contribute to oncogenesis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Prostate Cancer | TMPRSS2-ERG fusion leads to androgen-regulated overexpression of ERG, promoting invasion and proliferation. | PMID: 16204036, COSMIC |
| Ewing Sarcoma | EWSR1-ERG fusion (t(21;22)(q22;q12)) produces an oncogenic chimeric transcription factor. | PMID: 7987302, COSMIC |
| Acute Myeloid Leukemia | ERG overexpression and rearrangements (e.g., FUS-ERG) are associated with poor prognosis. | PMID: 16301605, COSMIC |
| Congenital Heart Disease | ERG haploinsufficiency contributes to cardiac defects in Down syndrome and 21q22 deletion syndromes. | PMID: 22921201, ClinVar |

Mutation functional classification
Loss of Function (LOF)
Loss-of-function mutations in ERG are rare; haploinsufficiency is associated with cardiac defects in 21q22 deletion syndromes.
Gain of Function (GOF)
Gain-of-function is primarily driven by gene fusions (TMPRSS2-ERG, EWSR1-ERG) leading to constitutive or overexpressed transcription factor activity.
Dominant Negative (DN)
Dominant-negative effects are not well characterized for ERG; fusion proteins may act as aberrant activators rather than dominant negatives.

Protein Summary
ERG is a 486-amino acid nuclear transcription factor belonging to the ETS family. It contains a conserved ETS DNA-binding domain that recognizes the core sequence GGAA/T. ERG regulates genes involved in hematopoiesis, vascular development, and cell adhesion. The protein is expressed in endothelial cells, hematopoietic stem cells, and certain epithelial tissues. Post-translational modifications include phosphorylation and sumoylation, which modulate its activity. In cancer, ERG fusions result in truncated or chimeric proteins that retain the ETS domain but lose regulatory regions, leading to aberrant transcriptional programs.
