Description

The ELL gene encodes a protein that functions as an RNA polymerase II elongation factor. It increases the catalytic rate of RNA polymerase II by suppressing transient pausing along the DNA template. ELL is also a component of the super elongation complex (SEC) and plays roles in transcriptional regulation, cell cycle control, and development. Chromosomal rearrangements involving ELL are associated with acute myeloid leukemia.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Acute Myeloid Leukemia (AML) Fusion of ELL with MLL (KMT2A) due to t(11;19)(q23;p13.1) translocation produces an oncogenic chimeric protein that deregulates HOX gene expression PMID: 8638120; COSMIC; ClinVar
Myelodysplastic Syndrome Similar MLL-ELL fusion implicated in therapy-related MDS PMID: 10477640
Solid Tumors (e.g., breast, colorectal) ELL overexpression or amplification may promote tumor growth via enhanced transcriptional elongation PMID: 23535663; COSMIC

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
t(11;19)(q23;p13.1) Translocation Rare Creates MLL-ELL fusion protein; drives leukemogenesis
c.1123C>T (p.Arg375Ter) Nonsense <0.1% Premature truncation; loss of elongation function
c.1456G>A (p.Glu486Lys) Missense <0.1% Unknown functional effect; reported in COSMIC
Mutation functional classification

Loss of Function (LOF)

Nonsense mutations (e.g., p.Arg375Ter) lead to truncated protein lacking elongation activity.

Gain of Function (GOF)

MLL-ELL fusion gains aberrant transcriptional activation properties, driving oncogene expression.

Dominant Negative (DN)

Not clearly documented for ELL.

Pathways

• RNA Polymerase II Transcription Elongation (Reactome R-HSA-75955)
• Super Elongation Complex (SEC) (Reactome R-HSA-8866654)
• MLL-ELL Fusion in Leukemia (KEGG hsa05202)

Protein Summary

The ELL protein (UniProt P55199) is 621 amino acids long and contains an N-terminal domain essential for elongation activity. It binds directly to RNA polymerase II and stimulates its processivity. ELL also interacts with other SEC components such as AFF1/4, ENL, and AF9. The protein is predominantly nuclear and is expressed in hematopoietic tissues, testis, and brain. Post-translational modifications include phosphorylation and ubiquitination, which may regulate its stability and function.
