Description

EIF4E3 (eukaryotic translation initiation factor 4E family member 3) encodes a protein that belongs to the EIF4E family of cap-binding proteins. These proteins recognize and bind the 5' cap structure of mRNAs, playing a key role in cap-dependent translation initiation. EIF4E3 is thought to modulate translation and may function as a tumor suppressor by competing with EIF4E for cap binding, thereby regulating the translation of oncogenic mRNAs.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (general) Dysregulation of cap-dependent translation; EIF4E3 may act as a tumor suppressor by competing with EIF4E. Limited; inferred from functional studies and expression data.
Acute myeloid leukemia (AML) Reduced EIF4E3 expression may contribute to leukemogenesis via altered translation of pro-survival mRNAs. Reported in some AML cohorts; further validation needed.

Protein Summary

The EIF4E3 protein (UniProt Q8N5X7) is a 25 kDa cap-binding protein with structural similarity to EIF4E. It binds the 5' cap of mRNAs but with lower affinity than EIF4E. EIF4E3 may regulate translation by sequestering cap structures or competing with EIF4E, thereby influencing the expression of specific mRNAs involved in cell growth and survival. Its expression is enriched in testis and hematopoietic tissues.
