Description

EIF3G encodes a subunit of the eukaryotic translation initiation factor 3 (eIF3), a multiprotein complex essential for translation initiation. The eIF3 complex binds to the 40S ribosomal subunit and promotes the formation of the 43S pre-initiation complex, facilitating mRNA recruitment and scanning. EIF3G specifically interacts with the eIF3 complex and is involved in stabilizing the complex and regulating translation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Dysregulation of eIF3G expression alters translation of oncogenes and tumor suppressors, promoting cell proliferation and survival. PMID: 25605274, COSMIC
Hepatocellular carcinoma Overexpression of EIF3G enhances translation of cell cycle regulators, contributing to tumor growth. PMID: 29367608
Breast cancer EIF3G upregulation correlates with poor prognosis and increased translation of pro-metastatic proteins. PMID: 31073040

Protein Summary

EIF3G is a 42 kDa protein that forms part of the eIF3 complex. It contains an RNA-binding domain and is thought to contribute to the structural integrity of the complex. The protein is ubiquitously expressed and plays a role in the recruitment of mRNAs to the ribosome. Dysregulation of EIF3G expression is associated with various cancers, where it may promote translation of growth-promoting mRNAs.
