Description

EIF3F encodes a subunit of the eukaryotic translation initiation factor 3 (eIF3) complex, which is essential for the initiation of protein synthesis. The eIF3 complex binds to the 40S ribosomal subunit and promotes the formation of the 43S pre-initiation complex. EIF3F is involved in regulating translation and has been implicated in cellular growth, proliferation, and apoptosis. Alterations in EIF3F expression or function are associated with various cancers and developmental disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal cancer Altered EIF3F expression affects translation of oncogenic mRNAs; loss of function may promote tumorigenesis COSMIC, NCBI Gene
Breast cancer EIF3F downregulation correlates with poor prognosis; may modulate translation of cell cycle regulators NCBI Gene, PubMed
Hepatocellular carcinoma EIF3F overexpression linked to increased proliferation and metastasis COSMIC, NCBI Gene

Mutation functional classification

Loss of Function (LOF)

Missense mutations (e.g., p.Glu278Lys) may impair eIF3 complex assembly or translation initiation activity.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported.

Dominant Negative (DN)

No evidence of dominant-negative effects.

Pathways

• Eukaryotic Translation Initiation (Reactome: R-HSA-72613)
• Cap-dependent Translation Initiation (Reactome: R-HSA-72737)
• Formation of the 43S pre-initiation complex (Reactome: R-HSA-72649)

Protein Summary

EIF3F is a 47 kDa protein subunit of the eIF3 complex. It contains a PCI domain (Proteasome, COP9, Initiation factor 3) that mediates protein-protein interactions within the complex. EIF3F is essential for stabilizing the eIF3 complex and facilitating mRNA recruitment to the ribosome. Post-translational modifications include phosphorylation, which may regulate its activity.
