Description

DYNLL1 encodes a dynein light chain protein that is a component of the cytoplasmic dynein motor complex. It functions as a dimeric adaptor that links cargo to the dynein motor for retrograde transport along microtubules. Beyond its role in motor transport, DYNLL1 participates in apoptosis by binding to and regulating pro-apoptotic factors such as BCL2L11 (Bim) and BMF. It also interacts with transcription factors and viral proteins, influencing cell cycle, gene expression, and viral pathogenesis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (multiple types) DYNLL1 overexpression or dysregulation alters apoptosis and cell migration, promoting tumor progression. ClinVar, COSMIC
Neurodegenerative disorders Impaired axonal transport due to DYNLL1 dysfunction may contribute to neuronal death. NCBI Gene, OMIM
Viral infections DYNLL1 binds to viral proteins (e.g., rabies virus P protein, HIV-1 integrase) to facilitate viral transport and replication. UniProt, NCBI Gene

Mutation functional classification

Loss of Function (LOF)

Mutations that impair dimerization or cargo binding reduce dynein motor efficiency and disrupt retrograde transport.

Gain of Function (GOF)

Not well documented; overexpression in cancers may confer a gain-of-function by enhancing cell survival.

Dominant Negative (DN)

Mutations that produce stable but non-functional dimers can inhibit wild-type DYNLL1 activity.

Gene Ontology (GO)

• microtubule-based movement • cytoplasmic dynein complex
• protein homodimerization activity • apoptotic process
• intracellular protein transport • binding

Pathways

• Intracellular transport via dynein motor
• Apoptosis regulation by BCL2 family members
• HIV-1 infection cycle

Protein Summary

DYNLL1 (LC8) is a highly conserved 10 kDa light chain protein that forms stable homodimers. It binds to the intermediate chain of cytoplasmic dynein and also interacts with a wide range of cargo proteins via a short linear motif. Its dimeric structure is essential for linking cargo to the motor complex. DYNLL1 also plays a role in apoptosis by sequestering pro-apoptotic BH3-only proteins, thereby modulating cell death decisions.
