Description

DUSP7 (dual specificity phosphatase 7) is a member of the dual specificity protein phosphatase subfamily. It inactivates MAP kinases (ERK, JNK, p38) by dephosphorylating both phosphothreonine and phosphotyrosine residues. DUSP7 is predominantly cytoplasmic and plays a role in cell proliferation, differentiation, and apoptosis regulation.

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.511C>T (p.Arg171Trp) Missense Rare Unknown functional impact

c.1045_1046insA Frameshift Rare Predicted loss of function
Mutation functional classification

Loss of Function (LOF)

Frameshift mutations (e.g., c.1045_1046insA) likely result in truncated, non-functional protein.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported.

Dominant Negative (DN)

Not described for DUSP7.

Pathways

• MAPK signaling pathway (KEGG: hsa04010)
• Negative regulation of ERK1/ERK2 cascade (Reactome: R-HSA-112409)

Protein Summary

DUSP7 encodes a 419-amino acid protein (UniProt Q16829) with a dual specificity phosphatase domain. It dephosphorylates phosphothreonine and phosphotyrosine residues within MAP kinases, thereby inactivating them. The protein is localized in the cytoplasm and is involved in fine-tuning MAPK signaling.
