Description

DUSP14 (dual specificity phosphatase 14) encodes a member of the dual specificity protein phosphatase subfamily. This phosphatase dephosphorylates phosphotyrosine and phosphoserine/threonine residues, negatively regulating MAP kinase (MAPK) signaling cascades, particularly ERK, JNK, and p38. DUSP14 is involved in immune regulation, cell proliferation, and apoptosis. It is also known as MKP6 (MAP kinase phosphatase 6).

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.314A>G (p.Tyr105Cys) Missense <0.01% Reduced phosphatase activity; associated with immune dysregulation
c.487C>T (p.Arg163Trp) Missense <0.01% Loss of function; impaired JNK dephosphorylation

Mutation functional classification

Loss of Function (LOF)

Missense mutations (e.g., p.Tyr105Cys, p.Arg163Trp) reduce catalytic activity, leading to sustained MAPK signaling.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in DUSP14.

Dominant Negative (DN)

Not described for DUSP14.

Protein Summary

DUSP14 is a 198-amino acid dual specificity phosphatase that dephosphorylates both phosphotyrosine and phosphoserine/threonine residues on MAP kinases (ERK, JNK, p38). It contains a conserved catalytic domain and a MAP kinase binding motif. The protein localizes to the cytoplasm and nucleus, and its expression is induced by stress stimuli. DUSP14 plays a key role in attenuating inflammatory and proliferative signals.
